I would think that it would make a big difference! You might start with Christopher Hogwood's recording of Bach's French Suites, Oiseau-Lyre 411 811-1. While no one is certain what tunings Bach actually used, these performances use six different temperaments that were common in Bach's time. I find the difference in sound immediately obvious; some of my friends can't hear it at all.
